Regulations between proteomes and metabolomes in response to oxidative stress

Redox states determine the fate of cells to differentiate, proliferate, defense, and die. An oxidative stress caused by the chemical, diamide, which induces protein thiol group oxidation. In monitoring the progress of oxidative stress alleviation, the S-thiolated proteins can be isolated by resin assisted method and detected by biotin switch techniques. Therefore, the protein redox states can be determined in the whole progress. On the other hand, the switch of metabolic pathway can also be observed by LC-ESI-MS. The replenishment of NADPH from metabolism should be important for reversing the oxidative stress that can be a model for studying more details and expansion of Warburg effect. In our preliminary results, several intermediates and the correspondent enzymes have been found to be critical for the redox and metabolism in the response of oxidative stress. The establishment of the model also helps in other situations, such as inflammation, differentiation, or infection.
